چکیده
Mononuclear copper(II) complexes based on the symmetric tridentate N3-donor ligands bis(pyridin-2-yl)methyl)amine (HBPA, L1) and its methylated counterpart, di(2-pyridylmethyl)amine (MeDPA, L2) have been prepared, characterized, demonstrated as functional models for lytic polysaccharide monooxygenase. These disrupt synthetic substrate, such p-nitrophenyl--D-glucopyranoside (-PNPG) into p-nitrophenol (PNP) D-allose via oxidative cleavage LPMOs do in nature. The observed spectroscopic kinetic analysis revealed that reaction proceeds hydroperoxide intermediate, whose electronic spectral signature has appeared at 350 nm absorption spectra with formation rate of 1.61 9.06 × 10-3 s1 respectively 1 2. Especially obtained product was newly 400 nm, indicates 7.52 5.45 s1.for 2 respectively. results affirm ability copper LPMOs.

A fast and sensitive activity assay for lytic polysaccharide monooxygenase
Background Lytic polysaccharide monooxygenases (LPMO) release a spectrum of cleavage products from their polymeric substrates cellulose, hemicellulose, or chitin. The correct identification and quantitation of these released products is the basis of MS/HPLC-based detection methods for LPMO activity. The duration, effort, and intricate analysis allow only specialized laboratories to measure LPMO...
متن کاملStructure of a Thermobifida fusca lytic polysaccharide monooxygenase and mutagenesis of key residues
Background Auxiliary activity (AA) enzymes are produced by numerous bacterial and fungal species to assist in the degradation of biomass. These enzymes are abundant but have yet to be fully characterized. Here, we report the X-ray structure of Thermobifida fusca AA10A (TfAA10A), investigate mutational characterization of key surface residues near its active site, and explore the importance of t...
متن کاملInteractions of a fungal lytic polysaccharide monooxygenase with β-glucan substrates and cellobiose dehydrogenase.
Lytic polysaccharide monooxygenases (LPMOs) are copper-dependent enzymes that catalyze oxidative cleavage of glycosidic bonds using molecular oxygen and an external electron donor. We have used NMR and isothermal titration calorimetry (ITC) to study the interactions of a broad-specificity fungal LPMO, NcLPMO9C, with various substrates and with cellobiose dehydrogenase (CDH), a known natural sup...
متن کاملStructure and boosting activity of a starch-degrading lytic polysaccharide monooxygenase
Lytic polysaccharide monooxygenases (LPMOs) are recently discovered enzymes that oxidatively deconstruct polysaccharides. LPMOs are fundamental in the effective utilization of these substrates by bacteria and fungi; moreover, the enzymes have significant industrial importance. We report here the activity, spectroscopy and three-dimensional structure of a starch-active LPMO, a representative of ...
متن کاملFungal cellulose degradation by oxidative enzymes: from dysfunctional GH61 family to powerful lytic polysaccharide monooxygenase family
Our understanding of fungal cellulose degradation has shifted dramatically in the past few years with the characterization of a new class of secreted enzymes, the lytic polysaccharide monooxygenases (LPMO). After a period of intense research covering structural, biochemical, theoretical and evolutionary aspects, we have a picture of them as wedge-like copper-dependent metalloenzymes that on red...
